The Aksaray Malaklisi originates from the high Anatolian plateau in central Turkey, an environment that presents a persistent physiological challenge: lower oxygen availability. The dog’s genome contains information detailing its adaptation to this hypoxic environment. This adaptation is primarily managed by the Hypoxia-Inducible Factor (HIF) pathway, a genetic system that regulates the body's response to oxygen levels.
Genetic studies in other high-altitude species, from Tibetan humans to their dogs, point to selection pressures on specific genes within this pathway. Key genes such as EPAS1 act as master switches, modulating red blood cell production to optimize oxygen transport without thickening the blood to dangerous levels. Other genes identified in high-altitude mammals include ARNT2 and THRB, both of which are part of the HIF system. The gene THRB, a thyroid hormone receptor, links oxygen regulation directly to the body's metabolic rate. Genomic analysis of the Aksaray Malaklisi indicates selection across these same genetic pathways, equipping this massive dog with a physiology tuned for life at elevation.
The genetics of a giant
The most apparent feature of the Aksaray Malaklisi is its immense size. Adult males often weigh over 85 kilograms (185 lbs) and stand 75 to 85 centimeters tall at the shoulder. This extreme size is not an accident; it is the result of intense artificial selection for dogs capable of defending livestock from large predators like wolves and bears. This selection has favored specific versions of genes that control growth.
Canine body size is heavily influenced by the growth hormone and insulin-like growth factor 1 (IGF1) pathways. Research comparing large and small dog breeds has identified several genes that drive extreme size. Variants in genes on the X chromosome, including Insulin Receptor Substrate 4 (IRS4) and Immunoglobulin Superfamily member 1 (IGSF1), are strongly associated with large dog breeds. These genes are involved in both growth hormone signaling and thyroid function, demonstrating a genetic link between the dog's metabolism and its massive frame. The dog's physical structure reflects its purpose. It possesses a large head, a muscular body, and famously pendulous jowls; the name "Malaklı" translates to "with lips." This powerful build, coded by its unique genome, makes it a formidable and effective flock guardian.
