A living fossil under threat
The tidal flats of the Yellow Sea host an animal that has navigated Earth's oceans for about 450 million years, predating most dinosaurs. The tri-spine horseshoe crab, Tachypleus tridentatus, is a species of marine arthropod that has remained morphologically similar for hundreds of millions of years. Despite surviving multiple mass extinction events, its populations are now in steep decline. The International Union for Conservation of Nature (IUCN) lists the species as Endangered, facing a high risk of extinction.
The primary drivers of this decline are human activities. South Korea's west coast, an important habitat, has experienced extensive coastal development and land reclamation, which has destroyed or degraded the sandy beaches and intertidal zones where the crabs spawn and juveniles mature. These shallow nursery areas are essential for the crab's life cycle; the larvae remain there for their first few years, molting multiple times as they grow. Pollution from industrial and agricultural sources also degrades water quality, further threatening their survival. In some parts of Asia, population declines have reached or exceeded 90% due to habitat loss and other pressures.
The price of blue blood
The endangerment of the horseshoe crab extends into human medicine. The animal's copper-based, blue blood contains cells called amebocytes. These cells are the sole source of a substance called Limulus Amebocyte Lysate (LAL), or Tachypleus Amebocyte Lysate (TAL) from Asian species. LAL has a unique property: it rapidly clots and forms a gel in the presence of endotoxins, which are harmful substances released by certain bacteria.
This reaction makes LAL an important tool for the pharmaceutical industry. It is used globally to test vaccines, injectable drugs, and implantable medical devices for bacterial contamination, ensuring they are safe for human use. To obtain it, hundreds of thousands of crabs are captured annually, have about a third of their blood extracted, and are then returned to the ocean. While the industry has historically claimed low mortality rates, studies suggest that between 10% and 30% of the bled animals die from the procedure. The practice also disrupts their mating cycles, as crabs are often harvested during their spawning season. A synthetic alternative, Recombinant Factor C (rFC), has been developed and is a viable replacement, but its adoption by the pharmaceutical industry has been slow. In 2023, South Korea's Ministry of Food and Drug Safety announced a phaseout of the horseshoe crab blood test in favor of synthetic alternatives.
